iSCORE-PD: Isogenic Stem Cell Collection to Research Parkinson’s Disease

iSCORE-PD Collection

The iSCORE-PD (Isogenic Stem Cell Collection to Research Parkinson’s Disease) collection is a set of 65 isogenic human embryonic stem cell (hESC) lines engineered to model genetic mutations associated with idiopathic Parkinson’s disease (PD). Developed under the Aligning Science Across Parkinson’s (ASAP) initiative, this resource supports research into the molecular and cellular origins of PD and facilitates the development of disease-modifying and potentially curative therapies.

All lines were derived from the well-characterized WIBR3 hESC line (RRID:CVCL_9767; NIH approval number NIHhESC-10-0079) and were extensively evaluated for pluripotency, culture stability, DNA damage response, differentiation potential, and genomic integrity.

Key aspects of the iSCORE-PD collection:

  • Isogenic control cell lines that are genetically identical to the edited lines but lack the specific Parkinson’s-associated mutation
  • Cell lines are engineered using CRISPR/Cas9 and prime editing to contain mutations in genes such as SNCA, PRKN, PINK1, LRRK2, and GBA1, among others

The iSCORE-PD collection was created through a collaboration among four laboratories:
